摘  要:为研究不同浓度IGF-β1、不同作用时间对体外培养的大鼠胸主动脉平滑肌细胞增殖的影响,并在此基础上探讨ERK信号转导通路在IGF-β1介导的大鼠血管平滑肌细胞增殖中的作用。取大鼠胸主动脉,用贴块法进行原代培养,取3~5代细胞。分别施加不同浓度IGF-β1(0、0.01、0.1、1、10ng/mL)作用24h和10ng/mL TGF-β1作用不同的时间(0、8、12、16、20、24h),用MTT法测定各组细胞吸光值,确定TGF-β1作用浓度和时间。再取生长正常的细胞,分四组:对照组,IGF-β1组,ERK阻断剂组和TGF-β1+ERK阻断剂组。继续培养24h后,用MTT法测细胞的增殖活性。TGF-β1浓度≥1ng/mL作用24h时吸光值明显低于对照组(P〈0.05),10ng/mL TGF-β1作用时间≥12h时吸光值明显低于对照组(P〈0.05),10ng/mL TCF-β1作用时间≥12h时吸光值明显低于对照组(P〈0.05);与对照组相比,其他三组吸光值均明显降低(P〈0.05),ERK阻断剂组吸光值低于TGF-β1组(P〈0.05)。TGF-β1呈浓度依赖和时间依赖方式抑制体外培养的血管平滑肌细胞的增殖,ERK信号转导通路可能不参与TGF-β1介导的大鼠血管平滑肌细胞增殖中的作用。To investigate effects of different TGF-β1 concentrations and action times on the proliferation of rat aortic smooth muscle cells in vitro and study the effects of extraeellular signal regulated kinase(ERK) pathway on the proliferation of rat vascular smooth muscle ceils induced by TGF-β1. The arota were removed, the primary VSMCs were isolated and cultured by the methods of sticking pieces. VSMCs of third to fifth cultured were used. The VSMC proliferation were detected after 24h with different concentrations of TGF-β1 (0,0.01,0.1.1.10ng/mL) and at different time points (0.8.12, 16,20.24h)with TGF-/3j treatment of 10ng/mL by MTT. Then VSMC normally growing were divided into four groups: control, TGF-β1 group, ERK blocking agent group and TGF-β1 + ERK blocking agent group. After cultured for 24h, the proliferation of VSMC were detected by MTT method. The extinction levels were lower than control when concentration of TGF-β1 was over 1ng/ml after 24h and active times were over 12h with TGF-β1 treatment of 10ng/mL( P 〈 0.05) ; A value of MTT assay in VSMC of other groups were decreased obviously than control group ( P 〈 0.05) and the extinction levels of ERK blocking agent group were lower than TGF-β1 group ( P 〈 0.05). Conclusion TGF-β1 exerts antiproliferative effects on vascular smooth muscle cells in a time-dependent and dose-dependent manner in vitro, It is possible that the ERK path- way does not participate the proliferation induced by TGF-β1.
