小檗碱对血管紧张素II诱导豚鼠结肠平滑肌细胞增殖的影响

摘　　要：目的观察小檗碱对血管紧张素II(AngII)诱导的培养豚鼠结肠平滑肌细胞(SMC)增殖的影响。方法组织贴块法培养的豚鼠结肠平滑肌细胞随机分成7组:对照组,模型组,小檗碱组(10-5mol/L、10-6mol/L、10-7mol/L),硝苯地平组和溶剂组。分别测定MTT(四甲基偶氮唑盐微量酶)吸收数值和细胞周期中各期细胞构成比。结果AngⅡ(0.1μmol/L)作用24h能使MTT吸收数值明显增加,并使SMC的G0/G1期细胞减少,细胞增殖指数增大。与AngⅡ组相比,AngⅡ加小檗碱(10μmol/L)组,AngⅡ加硝苯地平(10μmol/L),活细胞数减少,G0/G1期的细胞构成比增加,细胞增殖指数减少。结论AngII对SMC有促增殖作用,能被小檗碱所拮抗。Objective To investigate the effects of berberine and nifedipine on cell proliferation induced by Ang II in cultured smooth muscle cell of guinea pig colon. Methods Smooth muscle cell cultured by tissue-sticking method were randomly divided into control ,model, berberine （10^-5mol/L, 10^-6μmol/L, 10^-7 mol/L）, nifedipine ,and vehicle groups. MT T values and cell cycle distribution were measured. Results 24 h exposure to Ang Ⅱ （0.1 μmol/L ） caused an increase valube in MTT. Cell mitogenic activity was also increased. The proliferation was inhibited by berberine （10 μmol/L ）. Pretreatment with berberine increased percentages of G0/G1 period cells. Cells mitogenic activity induced by Ang Ⅱ were aslo reduced. Conclusion Berberine and nifedipine antagonized the proliferation of SMC of guinea pig colon induced by Ang Ⅱ.
