摘 要:目的研究HIV-1准种变化与人类白细胞抗原(human leucocyte antigen,HLA)限制性细胞毒性T淋巴细胞(cytotoxic Tlymphocyte,CTL)表位的相关性。方法从2对母婴传播HIV-1感染者血清内提取HIV-1核酸,进行逆转录-套式PCR(RT-nested PCR)扩增,产物克隆后,挑选阳性克隆行CSGE分析,对优势和劣势准种进行克隆测序分析,对氨基酸序列中可能的CTL表位进行分析。结果第1对感染的母子:母亲YJ体内存在的CTL表位在其54d的女儿WYF体内都存在,而WYF体内存在1株突变准种,其CTL表位在YJ体内不存在;第2对感染的母子:母亲CM和其12个月的儿子ZDB体内HIV-1env准种无相同的CTL表位。结论HIV-1在改变宿主后,随着时间的延长,HIV-1突变准种所导致HLA限制性CTL表位发生改变的概率增大。Objective To study the correlation of diversity of HIV-1 quasispecies and HLA-restricted CTL epitopes. Methods RNAs were extracted from the plasma samples of 2 pairs of individuals with HIV-1 vertical transmission (mother YJ and her 54-day daughter WYF, mother CM and her 12-month son ZDB) and polymersed by RT-nested PCR. PCR products were T-A cloned. Positive clones were analyzed by conformation sensitive gel electrophoresis (CSGE). The clones of major and minor quasispecies were sequenced. Sequencing resuits were analyzed to determine CTL epitopes. Results For the first pair mother-child, all CTL epitopes in env region of mother YJ' s HIV-1 quasispecies existed in env region of daughter WYF' s HIV-1 quasispecies, but one of CTL epitopes in env region of WYF' s HIV-1 minor quasispecies was not found in env region of mother YJ' s HIV-1 quasispeeies ; while for the second pair mother-child, no same CTL epitodes was found in env region of mother CM' s and son ZDB' s HIV-1 quasispecies. Conclusion The longer time after vertical transmission is, the more likely HLA restricted-CTL epitopes will change in mutated HIV-1 quasispecies.
