摘 要:目的观察严重烧伤早期不同时相点大鼠肠黏膜通透性的变化,探讨Rho激酶在这一过程中的作用及机制。方法常规制备烧伤大鼠模型后取回肠末段,分为正常对照组、烧伤组和烧伤6h+Y27632组,以荧光素标记法(FITC-D)检测大鼠肠黏膜在未烧伤、1、2、6、12、24h等不同时相点通透性的变化指标;用Westernblot法检测肠黏膜肌球蛋白轻链(myosinlightchain,MLC)、磷酸化MLC(p-MLC)和Rho激酶蛋白水平的变化。结果严重烧伤后大鼠肠黏膜通透性明显增加,并呈时相依赖性变化,6~12h达到最高;p-MLC以及Rho激酶蛋白表达均明显增加。上述蛋白表达的增强可在加入Rho激酶特异性抑制剂Y27632后消失。结论严重烧伤后肠黏膜Rho激酶激活和p-MLC表达增加,是导致其通透性增加及屏障功能损害的分子机制之一。Objective To observe the changes of intestinal permeability of severely burnt rat and investigate the role of Rho kinase in this process. Methods Fifty-six SD rats were included. Eight rats served as normal control. Burnt rat model was established with Rho kinase inhibitor Y27632. The changes in the other 48 rats, among which 8 rats were i.p. injected of intestinal permeability and the mucosa protein ROCK Ⅱ , myosin light chain ( MLC), holistic expression in 40 burnt rats were examined in the end-piece ileum at 1,2, 6, 12 and 24 h after burn, and in those of 8 rats in intervention group as well as in normal control. Results The intestinal permeability increased significantly in the severely burnt rats, and reached the highest in 6 - 12 h after burn. Phosphorylated MLC and ROCK Ⅱ increased obviously after severe burn, but Rho kinase inhibitor Y27632 counteracted this effect. Conclusion Rho kinase-mediated phosphorylation of MLC pathway is one of molecular mechanisms of increased intestinal permeability and impaired barrier function after severe burn.
