摘 要:目的通过观察内毒素休克模型大鼠肾组织中TLR2和TLR4及IκBαmRNA表达变化及人参二醇组皂苷(PDS)对其的影响,探讨内毒素引起肾脏损伤的机制。方法40只雄性Wistar大鼠随机分为对照组(control)、内毒素休克组(LPS)、地塞米松组(LPS+Dex)、人参二醇组皂苷低剂量组(LPS+PDSL)、人参二醇组皂苷中剂量组(LPS+PDSM)。舌下静脉注射内毒素(4 mg/kg)复制内毒素休克模型,4 h后测定肾组织中TLR2和TLR4及IκBαmRNA的表达。结果与对照组相比,LPS组TLR2和TLR4 mRNA的表达明显增加,IκBαmRNA的表达明显降低;而LPS+Dex、LPS+PDSL和LPS+PDSM组与LPS组相比,TLR2和TLR4 mRNA表达均降低,IκBαmRNA的表达增加。结论PDS能够下调肾组织中TLR2和TLR4mRNA表达,上调IκBαmRNA表达,具有减轻LPS引起肾脏天然性免疫反应损伤的作用。Objective To explore the molecular mechanism of nephridial tissue injury induced by endotoxin through observation of TLR2,TLR4 and IκBα mRNA expression of kidneys in endotoxic shock rats.Methods 40 Male Wistar rats were randomly divided into control group,lipopolysaccharide group(LPS),LPS+ dexamethasone group(Dex),LPS+panaxadiol low-dose group(PDSL),and LPS+panaxadiol middle-dose group(PDSM),respectively.TLR2,TLR4 and IκBα mRNA expression in testis were assayed 4 h after ranine intravenous injection of LPS(4 mg/kg).Results The TLR2 and TLR4 mRNA expressions in nephridial tissue of LPS groups were obviously higher than that of control group,but the IκBα mRNA expression was lower than that of control group.The TLR2 and TLR4 mRNA expression in nephridial tissue of LPS+Dex、LPS+PDSL and LPS+PDSM group were obviously lower than that of LPS group,but the IκBα mRNA expression was higher than that of LPS group.Conclusion PDS can lessen the kidneys injury induced by endotoxin through down-regulating TLR2 and TLR4 expression,up-regulating IκBα expression.
