摘 要:目的探讨CpG ODN在体外对人外周血单个核细胞的活性和肿瘤杀伤作用的影响。方法用CpG ODN刺激在体外分离培养的人外周血单个核细胞,检测其活性和对肝癌细胞系HepG-2杀伤作用。结果CpG ODN组PBMC培养上清IFN-γ显著高于对照组(P<0.05)。经CpG ODN刺激72小时后与HepG2细胞作用,48小时后乳酸脱氢酶在培养液中活力较对照组增强,差别具有显著性(P<0.05)。结论CpG ODN可明显提高人外周血淋单个核细胞对肿瘤的杀伤活性。Objective To discuss the antitumor effects of peripheral blood monouclear cells(PBMC)indueced by CpG ODN on hepatoma carcinoma cells HepG-2 in vitro.Methods PBMC was co-cultuered 96 well cell culture plates with CpG 2006,to detecte the IFN-γ concentration of each well's supernatant and co-culture the PBMC and hepatoma carcinoma cells HepG-2 for 48 hours,detect the LDH concentration of each mixed cell's supernatant to observe the injure of the hepatoma carcinoma cells HepG-2.Results PBMC induced by CpG ODN secrete more IFN-gamma than that of control group,the difference is obvious(P〈0.05),CpG ODN group made more injury to the hepatoma carcinoma cells than the control group.Conclusion PBMC induced by CpG ODN secrete more IFN-γ and appear more active to tumor cells.
