摘  要:目的测定莱菔硫烷(SFN)体内应用的生物半衰期。方法8只Wistar大鼠,腹腔注射SFN(3μmol/kg),分别于0.25、0.50、1.0、2.0、4.0、8.0h和16h从尾静脉和眼眶后静脉丛采静脉血400μL,采用Surveyor高效液相色谱仪测定SFN的血药浓度。结果血药浓度在30min左右到达高峰,后逐渐下降,利用MATLAB分别求出A=37.9817,k1=0.2304,k2=7.1973,作拟合曲线,求SFN在体内的消除半衰期t1/2=3.0078。结论大鼠体内注射SFN后,吸收快,消除快,合理用药间隔时间约3h。Objective To measure the biological half-life of sulforaphane (SFN) in vivo. Methods Eight Wistar rats were injected 3 μmol/kg SFN by intraperitoneal injection. 400 μL venous blood sample was collected at 1/4,1/2,1,2,4,8 and 16 hours after injection through vena caudalis or vena orbitalis posterior. The densities of SFN at different time phases were measured by high performance liquid chromatography (HPLC). Results The density peak of SFN in vivo was presented at 1/2 hour after intraperitoneal injection. The values of A,k1 and k2 were 37. 981 7,0. 230 4 and 7. 197 3 respectively. The elimination half life (t1/2) of SFN was 3. 007 8 according to the fitting curve. Conclusion SFN injected by intraperitoneal can be absorbed and eliminated swiftly in vivo. The reasonable interval of medication is about 3 hours.
