摘　　要：目的:构建人舌鳞癌(Tca-8113)细胞cDNA文库。方法:用Trizol法提取人舌鳞癌(Tca-8113)细胞的总RNA,使用Oligotex mRNA Mini Kit分离纯化mRNA,使用Library Construction Kit and cDNA SynthesisKit构建人舌鳞癌(Tca-8113)细胞cDNA表达文库。结果:人舌鳞癌(Tca-8113)细胞cDNA表达文库的初始文库滴度为5.4×105pfu/ml,重组率为95%,扩增后文库滴度为7.8×107pfu/ml。结论:成功构建了人舌鳞癌(Tca-8113)细胞cDNA表达文库,为进一步筛选人舌癌相关基因及蛋白奠定了基础。Objective To Construct cDNA Library from human tongue squamous cell carcinoma Tca-8113 cells. Methods Total RNA in human tongue squamous cell carcinoma Tca-8113 cells was extracted using Trizol reagent. The mRNA was isolated with Oligotex mRNA Mini Kit. The cDNA Library was constructed by Library Construction Kit and cDNA Synthesis Kit. Results The recombination rate was 95% and the titre of the primary library was 5.4×10^5pfu/ ml.The titre of amplified library was 7.8×10^7 pfu/ml.The length of most cDNA inserted in the library was 600-1 500bp. Conclusion Human tongue squamous cell carcinoma Tca-8113 cells is successfully constructed which provide subject for screening related genes and proteins of human tongue squamous cell carcinoma Tca-8113 cells.
