摘 要:以粉碎的大豆荚壳为材料,对总类黄酮提取条件和纯化方法进行研究。采用有机溶剂浸提法,就不同溶剂对类黄酮提取效果进行筛选,确定乙醇水溶液提取效果较好,并通过正交设计对大豆类黄酮提取条件进行了优化,确定了大豆类黄酮的最佳提取条件为φ=80%乙醇,提取温度为70℃,乙醇与大豆荚壳10∶1,回流提取24 h,提取率可达8.85%,显著高于次佳工艺。采用液-液分配分离得到了氯仿、乙酸乙酯、正丁醇类黄酮萃取物,并通过柱层析对萃取物进一步纯化,得到纯度为92.32%的总类黄酮提取液浸膏。因此,该提取与纯化方法对大豆荚壳类总类黄酮工业化生产具有一定的应用价值。The conditions for extracting and the method for purifying flavonoids from soybean hull were studied. Different organic solvents were used to screen out the extraction efficiency for total flavonoids, and the extraction conditions for total flavonoids were optimized by orthogonal design. The results are as follows: ethanol was the most effective in the selected solvents, the extracting temperature was 70℃, the ratio of 80% ethanol to soybean hull was 10: 1, the heating and refluxing time was 24 h. And the extractive ratio could be up to 8.85 %, which was significantly higher than that under the Hypo -optimal conditions. The total flavonoids dissolved in chloroform, ethyl acetate and butanol was proved by liquid -liquid separation, and the extracts were further purified by silica gel column chromatography, the purity of the total flavonoids could be up to 92.32%. So the conditions above have some application value in industrial processing of extracting total flavonoids from soybean hull.
