枸杞多糖对家兔动脉粥样硬化斑块中基质金属蛋白酶-2和血管内皮生长因子表达的影响  被引量:3

摘  要:目的:通过观察枸杞多糖(LBP)对兔动脉粥样硬化(AS)斑块中基质金属蛋白酶-2(MMP-2)和血管内皮生长因子(VEGF)表达的影响,来进一步探讨LBP治疗AS的作用及机制。方法:40只雄性新西兰兔随机分为4组:正常组、自然消退组、高剂量LBP组、低剂量LBP组,每组10只。正常组始终喂饲普通饲料;自然消退组及两组LBP组前10周均喂饲普通饲料+质量分数为2%胆固醇,在后10周,自然消退组喂饲普通饲料,高剂量LBP组与低剂量LBP组分别喂饲普通饲料+20mg/kgLBP(高剂量组)和10mg/kgLBP(低剂量组)。在第20周末,分别测量总胆固醇(Tch)甘油三脂(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C).第20周一次处死所有动物并观察主动脉形态学变化及应用免疫组织化学染色进行指标的检测及分析。结果:与正常组比,自然消退组Tch、TG、HDL-C、LDL-C明显升高(p<0.05);两组LBP组与自然消退组比较,Tch、TG、LDL-C明显降低(p<0.05),HDL-C明显升高(p<0.05);而高剂量LBP组与低剂量LBP组比较无明显差异。通过免疫组织化学染色发现,MMP-2和VEGF在AS斑块中呈阳性表达。对比于自然消退组,MMP-2和VEGF在两组LBP组中表达明显减少。结论:LBP不仅能够降脂,它还可以减少MMP-2和VEGF在AS斑块中的表达,这说明LBP能够对AS斑块性质产生影响,由此阻止动脉粥样硬化的发生发展。Objective :To investigate the effects of Lycium Barbarum Polysaccharide (LBP) on the expression of VEGF and MMP-2 in rabbit arteriosclerosis(AS) plaques for a better insight into the role and mechanism of LBP on the therapy of AS plaques. Methods: Forty healthy New Zealand rabbits were randomized into 4 groups: normal group, natural regressive group,high LBP group, low LBP group. There were 10 rabbits in ack group. Normal group always fed with normal diet; natural regressive group and LBP groups fed with normal diet + 2% cholesterol before 10 weeks. Natural regressive group only received normal diet after 10 weeks and LBP groups received normal diet + 20mg/kgLBP( high LBP group) or 10mg/kgLBP ( low LBP group). At the end of 20 weeks, total cholesterol ( Tch), tr/glyceride (TG), high density lipoprotein cholesterol ( HDL - C), low density lipoprotein cholesterol ( LDL - C) were measured, all rabbits were sacrificed. The morphological change of aorta was observed and further quantified using immunohistochemistry result. Results: Compared with nomal and natural regressive group, the LBP groupsTCh TG LDL - C lowered significantly( p 〈 0.05 ) ,HDL -C increased remarkably( p 〈 0.05 ), but the significant changes have not happened in high LBP group compared with in low LBP group. As found with SABC immunohistochemistry, VEGF and MMP- 2 were localized in AS plaques. Compared with natural regressive group, the LBP groups showed significantly lower level of VEGF and MMP - 2 expression. Conclusion: In addition to reduction of blood lipids, LBP appeared to markedly reduce the expression of VEGF and MMP -2 in AS plaques. It underlies its effection on the plaque quality and by which LBP can prevent the development of AS.
