B7-1基因转染儿童肝母细胞瘤苗抗肿瘤免疫作用的研究

摘  要:目的研究hB7-1基因以重组腺病毒载体转化儿童肝母细胞瘤后,转化细胞的抗肿瘤免疫作用。方法hB7-1基因重组腺病毒载体转入HepG2细胞后,获得高表达B7-1分子的HepG2/hB7-1细胞,MTT法检测HepG2/hB7-1细胞的生长情况,绘制生长曲线;ELISA法检测瘤苗体外对T淋巴细胞IFN-γ表达水平的影响。结果HepG2/hB7-1细胞表面的B7-1分于表达为18.5%,比HepG2细胞表达水平提高16.8倍。从HepG2/hB7-1细胞生长曲线可看到,转化hB7-1后细胞生长速度减慢,生长受到抑制。HepG2/hB7-1瘤苗刺激同种异体T淋巴细胞分泌IFN-γ的量明显超过HepG2细胞(P<0.01)。结论含hB7-1基因的重组腺病毒载体转染HepG2可以获得B7-1分子高表达的细胞株,降低肝母细胞瘤细胞的恶性表征,促进T淋巴细胞分泌IFN-γ,增强抗肿瘤免疫作用。Objective To assess the potential of immune-costimulating signal B7-1 vaccine in inducing anti-tumor immunity effect. Methods After HepG2 were transfected with B7-1 gene, the expression of B7-1 in cells and HepG2 cells growth were detected by flow cytometric analysis and MTT assay. The T cell function was detected by the method of ELISA. Results Flow cytometry revealed that HepG2 cells had a very low expression of B7-1 on the cell surface ( positive rate: 1.1% ). Conversely,the HepG2/B7-1 clone strongly expressed B7-1 ( positive rate: 18. 5% , increasing 16. 8 times compared to the former) on the cell surface. After transfection with B7-1 gene,the HepG2 cell proliferation slowed down, and the IFN-γ production by HepG2/B7-1 vaccine was higher than wild HepG2. Conclusion The transduction of B7-1 gene can enhance B7-1 expression in HepG2 cell, inhibit the proliferation of HepG2 cell, and activate T cell to pro- duce IFN-γ in vitro,hence enhancing the anti-tumor immunity.
