摘  要:采用单因子静态实验,考察了温度、NaOH和亮氨酸的摩尔比、L-亮氨酸浓度、树脂量对利用离子交换法制备L-亮氨酸螯合钙新工艺的影响,并测定了25℃下的离子交换动力学曲线。结果表明:交换可在室温下进行;NaOH与亮氨酸适宜的摩尔比1:1~1.3:1;L-亮氨酸浓度应控制在0.3 mol/L以下;适宜的树脂用量应满足树脂中钙与溶液中亮氨酸的摩尔比1:2;交换平衡时间为15 min。根据最佳工艺条件设计制备L-亮氨酸螯合钙的新工艺流程。采用小鼠对L-亮氨酸螯合钙和葡萄糖酸钙进行药动学和药效学实验对比,结果表明:L-亮氨酸钙的生物利用度比葡萄糖酸钙要高,其相对生物利用度是葡萄糖酸钙的175.3%,并具有缓释、药效时间长、不需要消耗胃酸等特点;L-亮氨酸螯合钙能显著性增加大鼠股骨重量,饲料中添加兼有促进小鼠体重增长的作用。Through the experiments of single factor, the effects of temperature, the ratio of NaOH to Leucine, the concentration of Leucine, the quantity of resin on exchange ratio were determined, its exchange kinetic curve at 25℃ was also measured. The results showed that exchange could be operated at room temperature, the proper mol ratio was between 1 : 1 and 1.3 : 1, the concentration of Leucine under 0.3 mol/L, the ratio of calcium in resin and Leucine in soltion was 1 : 2 during exchange, the equilibrium time was 15 minutes. Based on these conditions, the new technology of preparation of the complex of L-leucine Calcium was designed. The experimental comparisons of pharmacokinetics and pharmacodynamics between the complex of L-leucine calcium and glucose calcium were performed on the rats. The results showed that relative bio-availability of leucine calcium group to glucose calcium group was 175.3%, relative bio-availability of L- leucine calcium compound was higher than glucose calcium, and the complex of L-leucine calcium also had characteristics of being relaxed dosage and no consume of stomach acid, etc. The weight of rat was significantly increased in the calcium leucine compound group. Absorption effect of the complex of L-leucine calcium was better than glucose calcium at the same dosage.
