摘 要:目的探讨中药复方育泡饮对实验性自身免疫性卵巢早衰(POF)小鼠卵巢功能的影响。方法以成熟的SD雌性大鼠卵巢组织制备卵巢抗原,皮下多点注射免疫BALB/C雌性小鼠,建立卵巢早衰模型。在3次免疫后连续灌服中药复方,以强的松为阳性对照,酶联免疫法检测外周血雌二醇水平(E2)、卵泡刺激素(FSH)、外周血抗卵巢抗体(AoAb),IL-4、IL-10,观察卵巢的形态学变化,并计数各级卵泡及黄体数目。结果POF小鼠外周血E2下降,FSH升高,AoAb阳性,IL-4、IL-10明显提高,卵巢内有大量淋巴细胞浸润,各级卵泡及黄体数目减少。育泡饮能提高体内E2水平,减缓POF小鼠外周血FSH升高,抑制AoAb产生,降低IL-4与IL-10水平,减轻卵巢内淋巴细胞浸润,使各级卵泡及黄体数增加,育泡饮高剂量组比中剂量、低剂量组作用显著。结论育泡饮能减轻卵巢自身免疫性炎症损伤,对自身免疫性卵巢早衰具有治疗作用。Objective To discuss the influence of Yupao Decoction, a Chinese compound formula, on ovarian function of mice with experimental autoimmune premature ovarian failure (POF). Methods The ovarian antigen was elicited with ovarian tissue of mature SD female rats. The model of POF was established in BALB/C female mice by multiple sites subcutaneous injection. The mice were treated with Yupao Decoction after 3 times of injection. Prednisone was taken as the positive control. The levels of estradiol (E2), follicle- stimulating hormone (FSH), antiovarian antibody (AoAb), intedeukin-4 (IL- 4) and interleukin-10 (IL-10) in peripheral blood were determined by enzyme linked immunosorbent assay. The morphological changes of ovary were observed and the number of follicles in every stage and corpus luteum was counted. Results The level of E2 decreased, the levels of FSH, IL-4 and IL-10 increased, and AoAb was positive in peripheral blood of POF mice. The number of follicles in every stage and corpus luteum decreased. Infiltration of lymphocytes was also observed in the ovaries of POF mice. Yupao Decoction increased E2 level, reduced the increased FSH level, inhibited the production of AoAb,decreased the levels of IL-4 and IL-10, increased the number of follicles in every stage and corpus luteum, and reduced the infiltration of lymphocytes in the ovary. The efficacy was more significant in the group with high dose of Yupao Decoction than that in the groups with middle dose and low dose of Yupao Decoction. Conclusion Yupao Decoction can relieve the autoimmune inflammatory injury of the ovary, and shows a therapeutic effect on autoimmune premature ovarian failure.
