摘　　要：为了探讨HSV-2对血管内皮样细胞ECV304的致病特性,通过体外培养血管内皮样细胞ECV304并感染HSV-2后,采用RT-PCR和间接免疫荧光法检测HSV-2病毒的潜伏相关转录体基因的表达,并利用相差显微镜及电子显微镜观察ECV304感染HSV-2后的细胞形态变化。结果表明,血管内皮样细胞ECV304感染HSV-2后12～24h即可检测到潜伏相关转录体基因表达;感染后1～2d即可出现明显的细胞病变,提示HSV-2可感染ECV304细胞,并在其中活化复制,诱导ECV304细胞出现细胞融合和破裂死亡。To investigate the mechanisms for cytopathic effect （CPE） of herpes simplex virus type 2 （HSV-2） in ECV304 endothelial-like cells,RT-PCR and indirect immunofluorescence were used to detect HSV-2 infection. Phase-contrast and electron microscopy were performed to analyze morphology changes in infected and uninfected groups. In HSV-2-infected ECV304 cells, cytopathic effects were first observed approximately at 1-2 days post-infection. After 12-24 h infection, the majority of cells with CPE changes exhibited detachment from the monolayer,rounding of cells, cell shrinkage and membrane blebhing. The condensed chromatin and fragmented nuclei were examined and pronounced clustering of mitochondria with dramatic changes around nuclear was observed by electron microscopy,and on the second day, cell fusion was observed with weakened nuclear staining. ECV304 cells undergo predominantly necrosis after HSV-2 infection without obvious evidence of cell apoptosis.
