摘  要:目的:研究刺参糖胺聚糖对HepG2.2.15细胞HBsAg和HBeAg的抑制作用。方法:采用不同浓度的刺参糖胺聚糖作用于HepG2.2.15细胞,MTT法检测药物的细胞毒性,ELISA法检测培养上清中的HBsAg和HBeAg。结果:刺参糖胺聚糖对HepG2.2.15细胞分泌HBsAg和HBeAg的抑制作用存在时间和浓度依赖性。与对照组相比,给予8、4、2及1mg/mL4种不同浓度的刺参糖胺聚糖作用3、6d后,各药物处理组均能显著抑制HBsAg和HBeAg的分泌(P<0.01)。结论:刺参糖胺聚糖具有一定的抗HBV活性。Objective To study the effects of stichopus japomicus selenka Glycosaminoglycans (SJ-GAG) on hepatitis B virus (HBV) in vitro. Methods The human hepatoma(Hep)G2.2.15 cell line stably expressed hepatitis B virus particles in culture. The cells were exposed to different concentrations of SJ-GAG, respectively. Methyl, thiazolyl tetrazolium(MTF) assay was used to evaluate the cytotoxicity of the drugs. HBsAg and HBeAg were assessed by enzyme-linked immunosorbent assay (ELISA). Results The exposure of HepG2.2.15 cells to SJ-GAG resulted in a time-and concentration-dependent inhibition of HBsAg and HBeAg secretion. After exposed to four different concentrations of SJ-GAG for 3 and 6 days, the inhibition rates of extracellular HBsAg and HBeAg of each concentration increased significantly(P〈0.01 ). Conclusion SJ-GAG may be a new modality to treat hepatitis B infection.
