摘  要:为了研究薏苡属不同种质之间的关系,采用基因组原位杂交(GISH)技术,以四倍体栽培薏苡(2n=20,AABB)基因组总DNA作为探针,分别对广西的栽培薏苡、野生薏苡和水生薏苡体细胞中期染色体进行基因组原位杂交。杂交结果显示,栽培薏苡和野生薏苡染色体都被强烈而密集的杂交信号所标记,说明野生薏苡与栽培薏苡在基因组染色体水平上的同源程度很高,保守重复序列占很大比重;水生薏苡的基因组中有20条染色体的DNA成分与栽培薏苡的基因组DNA高度同源,推断供试的水生薏苡种属于广西六倍体水生薏苡居群。Genomic in situ hybridization (GISH) method was used to study the relationship of different Jobstears Coix species, and the cell metaphase chromosomes of Coix Lacryma-jobi L, Coix Agrestis Lour and Coix Aquatica Roxb were hybridized with labelled probes genome from Coix Lacryma-jobi L. (AABB genomes). The GISH results indicated that the chromosomes of Coix Lacryma-jobi L and Coix Agrestis Lout was labelled with strong and intensive hybridization signals, which showed that the homology between Coix Lacryma-jobi L and Coix Agrestis Lour genomes was very close at genomic chromosomes level, and the conservative repeated sequences accounted for a large proportion. Moreover, the DNA components of 20 chromosomes from Coix Aquatica Roxb genome was highly homologic with the genome DNA of Coix Lacryma-jobi L. It is concluded that testing Coix aquatica Roxb belongs to hexaploid Coix aquatica Roxb population in Guangxi.
