摘  要:【目的】研制检测麻疹IgG抗体的酶联免疫(EIA)试剂,用于确定人群免疫状况、考核麻疹疫苗接种效果及辅助诊断麻疹病毒的感染。【方法】以经超滤、PEG沉淀和蔗糖密度梯度超速离心纯化后的沪191株麻疹病毒为包被抗原,经试验确定EIA检测的最佳反应条件,最终建立麻疹病毒IgGEIA检测试剂盒,并与血凝抑制实验(HI)及SIGMA麻疹IgGEIA检测试剂盒作比较。【结果】纯化后沪191株麻疹病毒的比活性为775.76HAU/mg,以该纯化抗原制备的EIA检测试剂盒的检测下限为12.5mlU/ml,具有良好的线性(r=0.997)和重复性(CV为4.72%~13.51%)。4℃放置6个月、37℃放置3d,该试剂的灵敏度不变,说明其具有良好的稳定性。与血凝抑制实验(HI)相比,该试剂的灵敏度为99.46%,特异度为81.82%,一致性为97.58%,与SIGMA麻疹IgGEIA诊断试剂盒的一致率为95.12%。【结论】该检测试剂盒可以正确评价人体麻疹免疫状况,为流行病学调查提供了一个可靠、客观、灵敏和快速的手段。[Objective] To develope an enzyme immunoassay (EIA) diagnostic kit in detecting anti-measles IgG. [Methods] S191 measles virus was purified by ultrafiltration, PEG precipitation, density gradient ultracentrifugation and immobilized on polystyrene strip. The anti-measles IgG EIA diagnostic kit was established after determining the optimal reaction conditions by a series of tests. The kit was compared with hemagglutination-inhibitation (HI) test and SIG- MA anti-measles IgG EIA kit. [Results] The specific activities of purified S191 measles virus was 775.76HAU/mg . The sensitivity of the kit using S191 as coating antigen to detect anti-measles IgG was 12.5mIU/ml. The sensitivity, hn- earity (r = 0. 997) and reproducibility (CV: 4.72% - 13.51% ) were excellent, it remained the same quality even deposited at 4 ℃ for 6 months or at 37 ℃ for 3 days. As compared with HI test, the sensitivity was 99.46%, the sped- fieity was 81.82%, and agreement was 97.58%. The agreement with SIGMA anti-measles IgG EIA kit was 95.12 %. [Conclusions] The anti-measles IgG EIA kit can properly assess the condition of population immunization, It offers a credible, objective, sensitive and fast means in epidemiologieal investigation.
