摘 要:目的:通过研究白术对晚孕正常子宫平滑肌及经白细胞介素6(IL-6)处理的子宫平滑肌收缩的影响,以探讨白术对早产时宫缩的抑制作用。方法:①将人足月妊娠子宫平滑肌条浸没于恒温Tyrode灌流液中,通过张力换能器记录肌条的机械活动,直至稳定。②加入白术提取物后,观察不同浓度的白术对子宫平滑肌收缩的影响;③加入IL-6,观察不同浓度IL-6对子宫平滑肌收缩的影响;继而加入白术提取物后观察子宫平滑肌收缩的变化。结果:①不同浓度IL-6可引起子宫平滑肌收缩频率分别增快6.5%,12.4%,18.4%,振幅分别增强12.4%,26.4%,47.5%,并且呈剂量依赖关系(P<0.05)。②浓度为4 g/L和8 g/L的白术抑制正常子宫平滑肌收缩活动,使其振幅各下降32.5%、35.0%,频率各下降12.3%、13.2%(P<0.01),但无剂量依赖关系,且2 g/L的白术对其收缩无明显的作用(P>0.05)。③白术对加入IL-6导致子宫平滑肌的加强收缩亦有明显抑制作用。4 g/L和8 g/L的白术抑制其收缩活动,使其振幅各下降41.5%,41.5%,收缩频率分别减慢了17.9%,13.9%(P<0.01)。结论:白术对晚孕正常子宫平滑肌及对IL-6作用过的子宫平滑肌的收缩活动均有抑制作用,且对IL-6作用过的子宫平滑肌收缩活动的抑制作用强于对正常子宫平滑肌。实验结果表明白术对早产时的子宫收缩可能有作用,有望为开发中药治疗提供一定的理论依据。Objective. To investigate the effects of atractylodes macrocephala on the contraction of human preg- nant myometrium with or without interleukin-6 (IL-6). Methods: The myometrial smooth muscle seg- ments were divided into three groups: group A (Atractylodes macrocephala was added into Tyrode's solution), group B(IL-6 was added into Tyrode's solution) and group C (atractylodes macrocephala was added into Tyrode ' s solution after hypsiconcentration IL-6 had been added into Tyrode ' s solution). Results: IL-6 at 20, 40 and 80μg/L resulted in concentration-dependent contraction of uterine muscle (the frequency were respectively increased by 6.5%, 12.4% and 18.4%, the amplitude were respectively increased by 12.4%, 26.4% and 47.5%, respectively, P〈0. 05). When the concentration of atractylodes macrocephala were 4 and 8 g/L respectively, the contraction frequency were respec- tively inhibited by 12.3% and 13.2%, the amplitude were respectively inhibited by 32.5% and 35.0% (P〈0.01). There were visibly inhibitive effect of atractylodes macrocephala on the contraction of myometrial smooth muscle with IL-6 +4 g/L or IL-6 q-8 g/L Atractylodes macrocephala, the amplitude were respectively decreased by 41.5% and 41.5%, the contraction frequency were respectively decreased by 17.9% and 13.9% P〈0.01). Conclusion: Atractylodes macro- cephala was able to inhibit the contraction of human pregnant myometrium smooth muscle with or without IL-6. The inhibitive effect of the atractylodes macrocephala to the contraction with IL-6 was more powerful than without it. The study indicates that the atractylodes macrocephala has effects on the uterine contraction in premature delivery, and it also provideds some evidences for valuable application of atractylodes macrocephala.
