摘 要:目的研制几丁质微结晶分散体系,创造一种几丁质溶解环试验来替代传统的几丁质缺刻试验,用于鉴定细菌是否分泌几丁质酶,分解利用几丁质的一种新的实验方法。方法运用物理和化学综合手段,制备几丁质微结晶分散体系,几丁质浓度为4%,微结晶粒径〈0.8μm。采用制备的几丁质微结晶分散体系,进行几丁质溶解环试验和影响因子的研究。在几丁质培养基接种点周围出现明显的透明环,即为实验结果阳性,说明该细菌具有几丁质酶,对几丁质具有生物降解作用,反之为阴性。结果副溶血弧菌具有几丁质酶,能降懈、利用几丁质,而大肠杆菌则无此生物学功效。NaCl、pH、温度及菌量等因子对几丁质溶解环试验有直接的影响作用。结论几丁质溶解环试验是鉴定细菌能否分泌几丁质酶,并分解利用几丁质的一种直观、快速、准确的方法,在细菌学鉴定中具有实际意义。Objective To evaluate the dispersion system of chitin micro-crystal (DSCMC)for TCSC instead of the traditonal method. Methods DSCMC(4% chitin)was prepared by the physical and chemical methods.The diameter of chitin particle was〈0.8μm.Then the agar of chitin was prepared by DSCMC for TCSC. The relationship between result of TCSC and influences Factor was researched. TCSC positive:The obvious transparent circles were appeared surrounding of inoculating point in chitin agar,which meant bacteria could decompound chitin and secrete enzyme of chitin. TCSC negative:The transparent circle was not appeared surrounding of inoculating point chitin agar. Results The results of TCSC could be influenced directly by NaCl,pH,temperature and bacterial quantity.Conclusion TCSC is a new method to determinate bacterial ability of chitin decompounding.This new method is more intuitionistic,exacter and faster than the traditional method.
