摘  要:目的:观察海洋低温溶菌酶对体外培养的人脐静脉内皮细胞超微结构的影响,探讨海洋低温溶菌酶抗肿瘤血管形成的机制。方法:体外培养人脐静脉内皮细胞,用不同浓度的海洋低温溶菌酶处理24h后,利用透射电镜观察细胞超微结构的变化。结果:海洋低温溶菌酶处理后人脐静脉内皮细胞出现超微结构变化,包括核内染色质浓缩及边集,胞质内细胞器肿胀和空泡化,呈现典型的细胞凋亡改变。这种改变呈剂量依赖性。结论:海洋低温溶菌酶可引起人脐静脉内皮细胞损伤,导致细胞凋亡,从而可能诱发其抗肿瘤血管生成的作用。Objective:To observe the effects of marine low-temperature lysozyme on the ultrastructure of human umbilical vein endothelial cell (HUVEC) in vitro and to investigate its mechanism of inhibitory effect on tumorous angiogenesis. Methods: HUVEC was cultured in vitro for 24h and then treated with marine low-temperature lysozyme of different concentrations(200,400 and 800 μg/mL) for 24 h. The uhrastructural changes of HUVEC were observed under the transmission electron microscope. Results:The uhrastructure of HUVEC was obviously changed after treated with marine low-temperature lysozyme and showed various stages of chromatin condensation, including initial condensation, migration of chromatin at nuclear membrane, complete condensation of nucleus and vacuolation of cytoplasmic organelles. These typical apoptotic changes also showed a concentration-dependent effect. Conclusion: Marine low-temperature lysozyme can damage and induce apoptosis of HUVEC and it might inhibit the growth of tumor by its inhibitory effect on the angionesis.
