凉血化瘀方对蓝光诱导人视网膜色素上皮细胞凋亡的影响

摘　　要：目的探讨唐由之研究员经验方凉血化瘀方对蓝光诱导体外培养的人视网膜色素上皮（retinal pigment epithelium，RPE）细胞凋亡的影响。方法将RPE细胞分为四组，用波长为470．500nm，光照强度在（2000±500）LUX的蓝光照射12小时，处理组分别于光照前30分钟加入不同浓度的凉血化瘀方中药粗提物及100umot／L的维生素C，终止光照后用，用荧光素标记的连接素V／碘化丙锭（Annexin V—fluorescein isothiocyanate／Propidium iodium，Annexin V—FITC／PI）双染色法，上流式细胞仪检测，观察细胞凋亡的情况及不同浓度的凉血化瘀方中药粗提物对蓝光诱导RPE细胞凋亡的影响。结果蓝光照射体外培养人RPE细胞后，引起的细胞变化以细胞凋亡为主，光照前加入不同浓度的凉血化瘀方中药粗提物及维生素C后，凋亡细胞数量明显减少，且凉血化瘀方组凋亡细胞数量少于维生素C组。结论凉血化瘀方对蓝光诱导体外培养的人视网膜色素上皮凋亡具有保护作用。OBJECTIVE To observe the effect of Liangxue Huayu decoction to blue light - induced apoptosis in cultured retinal pigment epithelial（RPE）cells of human. METHODS Human RPE cells were divided into 4 groups. Respectively, group A was the normal control group, group B was the blue light damage group, gruopC was the cells that had 100umol/L VitC, group D was the one that RPE cells had different concentration of Liangxue Huayu decoction. RPE cells of all groups were exposed to blue light with the same intensity, same time of illumination, same finish time of the culture. The apoptosis of RPE cells was observed by Annexin V fluoresein isothiocvanate（ FITC）/propidiumiodide（PI） flowcytom- etry. RESULTS The human RPE cells were exposed to blue light, and the main influence to the cells was apoptosis. The quantity of apoptosis RPE cells of group B is the greatest. The quantity of apoptosis cells of the other groups which were cultured with VitC, or liangxue Huanyu decoction, is less than groupB, but still more than group A. The protective effect of Liangxue Huayu decoction is superior to VitC. CONCLUSIONS The protective effect of Liangxue Huayu decoction is confirmed to in vitro human RPE cells＇ injury, which is mainly apoptosis, induced by lllumination with blue light.
