摘 要:目的研究竹节人参提取物对大鼠肾缺血再灌注损伤的影响。方法建立肾缺血再灌注损伤大鼠模型,32只大鼠分为治疗组(n=8)和模型组(n=16)。另设假手术组(n=8)作为对照。治疗组于肾动脉夹闭时给予竹节人参提取物0.3 g/kg静脉注射,再灌注时又给予原药量的1/2;模型组同时给予等量0.9%氯化钠溶液静脉注射。另取仅作麻醉、开腹,不阻断肾血流的8只大鼠作为对照组。观察肾组织中P-选择素、细胞间粘附分子(ICAM-1)的表达及丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性变化,检测血清尿素氮(BUN)和肌酐(Cr)的含量。结果缺血再灌注后,模型组大鼠肾组织中P-选择素、ICAM-1明显表达,MDA含量增高,SOD活性下降,BUN和Cr含量明显上升;治疗组肾组织中P-选择素、ICAM-1表达受到抑制。相对模型组MDA含量降低,SOD活性增高,BUN和Cr含量降低。结论P-选择素和ICAM-1参与肾缺血再灌注损伤机制,竹节人参提取物明显减轻脂质过氧化反应,对肾缺血再灌注损伤大鼠具有保护作用。Objective To investigate the effect and protective effect of Panaxjaponicus C. A. Meyer on renal ischemia - reperfusion injury in rats. Methods Rat models of renal ischemia - reperfusion were established, and 32 male rats were randomly divided into 3 groups, treatment group with Panaxjaponicus C. A. Meyer (n =8), renal ischemia - reperfusion injury model group (n = 16) and sham operation group (n =8) as the control. Panaxjaponicus C. A. Meyer (0.3g/kg) was given intravenously during clamping renal artery in treatment group and one half of the dose was infused after opening the clamp during reperfusion. In model group equal amount of O. 9% sodium chloride solution was given. In the sham operation group (control group), the rats were anesthetized with laparotomy but without blocking renal blood flow. The P - selectin and ICAM - 1 expression, malonyldialdehyde (MDA) contents and super oxide dismutase(SOD) activi- ties in renal tissues, level of serum blood urea nitrogen(BUN) and creatinine(Cr) were detected. Results After renal ischemia reperfusion injury, there were significantly up - regulated expressions of P- selectin and ICAM -1 ,with increased MDA content and declined SOD activity in renal tissue, and significant increase in BUN and Cr content in model group, and these effects in treatment group were significantly depressed as compared with model group. Conclusion It is thought that the adhesion molecules P - selectin and ICAM - 1 take part in the process of renal ischemia - reperfusion injury, and Panax japonicus C. A. Meyer significantly reduce the re- action of lipid peroxidation and has a protective effect on renal ischemia - reperfusion injury.
