GM-CSF与SS融合表达质粒对小鼠淋巴细胞增殖及GH和IGF-1分泌的影响

摘　　要：以小白鼠为试验对象,分别肌肉注射生理盐水及pcS/2SS、pGM-CSF/SS、pGM-CSF+pcS/2SS质粒,每只50μg,免疫后第3周以相同剂量加强免疫1次,测定不同质粒对小鼠淋巴细胞增殖反应及血浆生长激素(GH)、胰岛素样生长因子-1(IGF-1)分泌水平,探讨GM-CSF对生长抑素DNA疫苗的免疫增强作用。结果表明:GM-CSF可增强小鼠脾脏淋巴细胞的增殖反应,以pGM-CSF+pcS/2SS组的增殖能力最强,显著高于pcS/2SS组和生理盐水组;同时注射2种表达质粒(pGM-CSF+pcS/2SS)的GH分泌水平和融合表达质粒(pGM-CSF/SS)的IGF-1分泌水平明显高于生理盐水组和pcS/2SS免疫组。这证明GM-CSF对生长抑素DNA疫苗有一定的免疫增强作用,可促进生长抑素DNA疫苗的免疫反应,提高血浆GH和IGF-1的分泌水平。A total of 40 mice at the age of 22 days was divided into 4 groups to study the immuno-enhancement of genetic adjuvant GM-CSF on somatostatin （SS） DNA vaccine. Ten mice in each group were given tWO intramuscular injections at interval of 2 weeks with saline solution as control （group 1）, pcS/2SS （group 2）, pGM-CSF/SS （group 3）, and pGM-CSF＋pcS/2SS （group 4）, respectively. The results showed that lymphocyte proliferation response of pcS/2SS along with pGM-CSF was higher than that of the others, indicating that GM-CSF possesses an immuno-enhancement. Moreover, the GH level in sera of pcS/2SS along with pGM-CSF and IGF-1 level in sera of pGM-CSF/SS were higher than that of other groups, showing that the levels of GH and IGF-1 begin to rise at the 2nd and 4th week of primary immune, respectively. The data suggested that genetic adjuvant GM-CSF can enhance immune response of somatostatin DNA vaccine and increase GH and IGF-1 level in sera of mice.
