摘 要:目的探讨发形霞水母毒素(Cyanea capillatafull venom,CFV)的体外溶血活性及其影响因素。方法超声破碎法分离得到CFV,测定其半数溶血分数(HU50),探讨温度、pH、放置时间对溶血活性的影响,并进一步研究不同动物红细胞对溶血活性的敏感性。结果CFV溶血活性的HU50为5.82μg.mL-1;溶血活性对温度和pH变化耐受性差,在40℃和pH 8.0时有最大溶血活性;25℃和37℃放置12 h CFV溶血活性迅速下降至18.6%和17.4%;对不同动物红细胞敏感性差异较大,其中以家犬红细胞最为敏感。结论CFV有显著的溶血活性。Objective To investigate the factors influencing hemolytic activity of venom from the jellyfish Cyanea capillata. Methods The HU50 of Cyanea capillata full venom (CFV) was obtained by dose-response curve of hemolytic activity, then some factors such as pH, temperature, time and different animals erythrocytes were given to observe the hemolytic activity changes of CFV. Results The HU50 of CFV against SD rat erythrocytes was 5.82 μg · mL^-1. CFV had maximum hemolytic activity at the pH 8. 0 and the hemolytic ratio was 0.93. Hemolytic activity was temperature-sensitive and reduced sharply when CFV was preincubated at temperature over 40 ℃. With the passage of time, the hemolytic activity of CFV reduced in 25 ℃ and 37 ℃. Hemolytic activity displayed obvious changes with different animal erythrocytes and CFV had maximum hemolytic activity to the dog erythrocytes. Conclusion CFV displayed remarkable hemolytic activity in erythrocytes.
