摘  要:目的观察枇杷叶三萜酸(TAL)对慢性支气管炎(CB)大鼠肺泡巨噬细胞(AM)一氧化氮合酶(iNOS)和一氧化氮(NO)生成的影响,探讨其对慢性支气管炎抗炎作用的部分机制。方法应用卡介苗联用LPS的方法复制大鼠CB模型,观察TAL对CB大鼠AM iNOS表达、活性及NO释放的影响。结果在卡介苗联用LPS诱导大鼠CB模型中,TAL能降低AM iNOS的mRNA及蛋白的表达,并且抑制NO的释放。结论TAL抑制CB大鼠AM iNOS的表达及其活性,从而减少NO的释放可能是其对慢支的治疗与防治作用机制之一。Objective To investigate the effect of triterpene acids of eriobotrya japonica (Thunb.) lindl, leaf (TAL) on the iNOS and the production of NO in alveolar macrophages (AM) of chronic bronchitis (CB) rats and the mechanism related to anti-inflammatory action. Methods The model of CB was established by using the BCG plus LPS method. The effect of TAL on the expression of iNOS and the production of NO was measured by immuo- cytochemistry and colorimetric assay. Results TAL(big/moderate dose group) could reduce the release of NO obviously in AM of CB rats. It could also lower the average optical density of iNOS histoehemistry as well as the iNOS mRNA expression by RT-PCR in AM of CB rats. Conclusion TAL can reduce the expression of iNOS and release of NO in AM of CB rats, it may be the one of the mechanism of the preventive and therapeutic effect in chronic bronchitis.
