蒸汽对烟草漂浮苗干根中烟草花叶病毒的失活效果

摘　　要：为了评价蒸汽用于烟草漂浮育苗旧苗盘残留物中烟草花叶病毒(TMV)的消毒效果,采用枯斑寄主和K326接种方法,测定了蒸汽对烟草漂浮苗花叶病株干根中TMV的失活效果。结果表明,96℃蒸汽蒸3～60 min,对病苗干根中TMV失活效果高,可达99.9%。血清学检测结果表明,蒸汽消毒3 min能有效破坏病根样品中TMV抗原。推荐旧苗盘TMV蒸汽消毒方法为:苗盘润湿后96℃蒸汽保持不少于3 min。但在96℃蒸汽中保持5 min后,普通聚苯乙烯泡沫苗盘在冷却过程中变形。In order to evaluate inactivation effects of steam to tobacco mosaic virus （TMV） in residue of float tray, the inactivation effects to TMV in dry roots of tobacco float seedling was determined by local lesion assay and inoculation to tobacco variety K326. The results indicated that steaming at 96℃ in 3 - 60 minutes could get evident inactivation to TMV in dry roots of tobacco seedling, with inactivation ratio of 99.9%. TAS-ELISA and TMV-Imunostrip test showed that steaming in 3 minutes could fully destroy TMV antigen in dry roots. Steaming at 96℃ in more than 3 minutes is recommended to apply in disinfection of floating tray. But foamed polystyrene plastic tray is apt to distort after steaming at 96℃ for 5 minutes.
