赖型钩体毒力相关基因OmpL17表达纯化及其产物的趋化作用

摘　　要：目的研究钩体017株外膜蛋白新基因ompl17与钩体肺大出血的相关性。方法提取钩端螺旋体017株基因组DNA,扩增出ompL17基因,与表达载体pGEX-4T-1重组,诱导表达OMPL17蛋白,然后纯化得到活性表达产物,并建血管基底膜,分析该产物的趋化活性。结果SDS-PAGE和Western-blot分析证实诱导表达并纯化得到OMPL17蛋白,趋化作用分析证明该基因表达产物具有趋化活性。结论成功表达纯化出ompL17基因的蛋白,观察到该基因表达产物具有趋化活性,为赖型钩体的分子机制的进一步阐明奠定了基础。To analyze a new gene ompL17＇ relativity to the PDH of Leptospira. Methods ompL17 gene was cloned, expressed and purified. And the chemotaxis function of Ompl17 protein to blood cells was observed through constructing blood vessel base membrane. Results SDS-PAGE, Western-blot certified the successful expression and purification of the gene ompL17. Ompl17 protein had chemotaxis function to blood cells. Conclusion The result of this study lays foundation for further exploring molecule pathogenesis of Leptospira interrogens serovar lai.
