摘 要:目的探讨脂多糖(Lipopolysaccharide,LPS)诱导人肺泡上皮细胞(A549)一氧化氮(Nitric oxide,NO)分泌的影响。方法对A549给予不同浓度的LPS,分别收集其培养上清液,用硝酸还原法检测NO浓度。结果不同浓度LPS诱导A549,NO分泌量高于未经LPS诱导的对照组,且具有浓度、时间依赖性。结论LPS诱导A549NO的分泌,提示LPS具有诱导肺泡上皮细胞急性炎症和氧化应激的作用。Objective To investigate the effects of quercetin on nitric oxide (NO) in human pulmonary epithelial cells (A549) induced by lipopolysaccharide (LPS). Methods The supernates of LPS-induced cultured A549 from different intervention groups were collected to measure the concentration of NO by nitrate reductase assay. Results The concentration of NO in supernates of LPS-induced A549 was higher than that in the control group (P〈0. 05) in a dose- and time-dependent manner. The concentration of NO in supernates of quercetin intervention groups at 10, 15μmol/L was lower than that in non-intervention group (P〈0. 01), but that at 5μmol/L was no difference (P〉 0.05). Conclusion Quercetin can decrease LPS-induced NO secretions in A549, which suggests that quercetin has anti- inflammatory and anti-oxidant stress properties on A549.
