SLC11A1基因多态性在新发和复发肺结核患者中的分布

摘　　要：目的探讨SLC11A1基因多态性在中国汉族人群新发和复发肺结核患者中的分布。方法在中国汉族人群中,选择新发肺结核患者、复发肺结核患者和健康对照者各30例,应用SNaPshot SNP分型技术检测以上人群SLC11A1基因多态性位点INT4、D543N和3′UTR的基因型,采用Logistic回归分析和2χ检验进行统计学处理。结果病例组3′UTR位点TGTG+/del基因型频率显著高于对照组(P=0.038),病例组与对照组INT4和D543N位点各基因型频率无显著性差异,病例组D543N和3′UTR位点联合分析显示GG/+del及GA/+del基因型频率均显著高于对照组(P=0.048和P=0.034)。新发肺结核患者与复发肺结核患者3′UTR和D543N/3′UTR基因型频率无显著性差异。结论SLC11A1基因3′UTR位点多态性和D543N/3′UTR单倍体型可能是中国汉族人群肺结核发病的易感因素,但与肺结核复发的关系有待进一步研究。Objective To investigate the distribution of solute carrier family 11 member 1 protein （SLC11A1 ） gene polymorphisms between new and recrudescent cases of pulmonary tuberculosis （PTB） in Chinese Han population. Methods Thirty new cases, thirty recrudescent cases of PTB and thirty healthy controls of Chinese Han population were enrolled in the study. SNaPshot SNP genotyping technique was used to type the polymorphisms of SLC11A1 gene,which at sites of variants intron 4（INT4） ,D543N and 3＇ untranslated region（3＇UTR）. All the data were analyzed by logistic regression analysis and chi-square test. Results TGTG ＋/del genotype at 3＇UTR site occurred more frequently in patients than in controls（ P = 0. 038 ）. No significant differences were observed in the distribution of at genotypes INT4 and D543N sites between pa-tients and controls. Combined analysis of D543N and 3＇UTR variants demonstrated that GG/＋ del and GA/ ＋ del genotypes occurred more frequently in patients than in controls （ P = 0.048 ＆ P = 0.034）. No significant differences were observed in the distribution of genotypes at 3 ＇UTR and D543N/3 ＇UTR between new and recrudescent cases. Conclusion SLCllA1 gene polymorphisms in 3＇UTR and heterozygosis of D543N and 3＇UTR variants might be susceptible to tuberculosis in Chinese Han population, but the relationship between SLC11A1 gene polymorphisms and recrudescence of PTB should be further studied.
