摘 要:采用尼龙线栓塞大鼠大脑中动脉建立局部性脑缺血模型,用盐酸非那嗪奈(1)的低、中、高剂量组(0.5、1、2mg/kg)、尼莫地平组(2mg/kg)和生理盐水组分别在脑缺血后1、3、6h给药,缺血24h后观察各组对大鼠脑梗死区比例和神经行为学缺陷的改善作用。结果表明,与生理盐水对照组相比,1及尼莫地平在脑缺血后1h给药均可使脑坏死区比例显著降低,动物行为学评分明显改善。与对照组相比,1中、高剂量组在3个给药时间点均有明显疗效,低剂量组在脑缺血后6h给药无显著性差异。Focal cerebral ischemia model was established by using nylon thread occlusion of the middle cerebral artery of rats. The low, medium and high doses of fenazinel dihydrochloride (0.5, 1, 2mg/kg), nimodipine (2mg/kg) and saline were administered after ischemia for 1, 3 and 6h. The therapeutic effects on the neurological defects and the ratio of the areal fraction of cerebral infarction after ischemia for 24h were investigated. The results showed that compared with the saline, fenazinel dihydrochloride and nimodipine administered after ischemia for lh could significantly reduce the areal fraction of cerebral infarction and improve the neuroethological evaluation. The therapeutic effects of the middle and high dosage groups of fenazinel dihydrochloride administered after 1, 3 and 6h were significantly better than those of the saline group. The low dosage group of fenazinel dihydrochloride administered after 6h had no significant difference with the saline group.
