摘  要:目的:探讨简便、快捷、灵敏的观察阴茎白膜损伤愈合过程中胶原类型、分布动态变化的方法。方法:在15只新西兰兔阴茎背侧切除白膜5mm×5mm造成缺损,分别在术后2,6,12周(每次5只)取材,苦味酸天狼星红染色,偏振光显微镜观察。结果:术后2周,胶原纤维纤细,由Ⅰ型和Ⅲ型胶原纤维共同组成,此后,Ⅰ型胶原纤维逐渐增多,Ⅲ型胶原纤维逐渐减少,24周时Ⅰ型胶原纤维为主,Ⅲ型已不明显,不同时间点同型胶原纤维含量无显著性差异(P>0.05),术后2周,损伤白膜局部Ⅲ型胶原纤维占大多数;术后6周,术区Ⅰ型胶原纤维明显增加,但排列尚未有序;术后12周,Ⅰ型胶原纤维增粗、增多,呈斜行螺旋状交叉排列。结论:苦味酸天狼星红偏振光不仅能区分Ⅰ、Ⅲ型胶原的类型还能清晰地显示Ⅰ、Ⅲ型胶原的形态、分布和比例关系,具有操作简单、省时、花费少,而且特异性强、灵敏度高。Objective: To find a convenient, swif and sensitive method for observation of the developing change of types and distribution of collagen during healing process of albuginea trauma. Methods:15 male New Zealand white rabbits underwen excision of an ellipse and a 0.5 cm × 0. 5cm defect was made in notal side of tunica albugineand,The animals were euthanized and specimens were harvested at intervals of 2,6 and 12weeks,The specimens were stained by picrosirius and observed by polarimicroscope. Results:Collagen Ⅰ and Ⅲ occupied mostly the trauma site 2 weeks postoperatively;collage Ⅰ increased collagen Ⅲ decreaced evidently at the 6 weeks ,but its array was still not regular;collagen Ⅰ was not only thicker but also much more in original defect, and collagen Ⅰ occupied mostly the grafting site at12weeks. Conclusion,, Picrosirius stain can clearly display the distribution,form and proportion of collagen Ⅰ and Ⅲ. This method is simple,convenient,cheap, specific and highly sensitive.
