阿魏酸钠对多囊卵巢大鼠颗粒细胞凋亡的影响

摘　　要：目的:研究阿魏酸钠(SF)对多囊卵巢大鼠体重、血清激素和卵巢颗粒细胞凋亡的影响。方法:20只SD雌性大鼠注射脱氢表雄酮(DHEA)建立多囊卵巢大鼠(PCO)模型,随机分为SF组和DHEA组,观察用药前后动物体重和血清激素水平变化情况;TUNEL法和免疫组化法检测卵巢颗粒细胞凋亡及凋亡相关蛋白Bcl-2和Bax表达水平变化。结果:SF组血清T水平和E2浓度均较DHEA组降低(P<0.05),血清P浓度则明显增加(P<0.05);DHEA组镜下可见卵巢颗粒细胞层数明显增加至4-8层,细胞凋亡率显著低于SF组(P<0.001),其Bcl-2表达较空白组低(P<0.05),Bax表达则较高(P<0.001),Bcl-2/Bax的比值下降;SF组Bcl-2的表达较DHEA组高(P<0.001);Bax表达较低(P<0.001),Bcl-2/Bax的比值增加。结论:阿魏酸钠改善多囊卵巢大鼠内分泌状况可能与调节Bcl-2和Bax蛋白表达,发挥其抗细胞凋亡有关。Objective To investigate the effects of sodium ferulate（SF） on weight ,serum hormone level,and the apoptosis of ovarian granulosa ceils of rats with polycystic ovary（PCO）. Methods Rat PCO model was established by dehydroepiandrostone（DHEA） injection and randomly divided into SF groups and DHEA group. The change of weight and serum hormone levels of PCO rats was observed before and after soduium ferulate treatment. TUNEL assay was used to detect the apoptosis of ovarian granulosa cells. Immunohistochemical analysis was used to demonstrate the expressions of the Bcl-2 and Bax in ovarian granulosa ceils. Results The increase range of weight was significantly decreased in SF group than that in DHEA group （ P 〈 0.001 ）, Both testosterone and estradiol level were significantly lower in SF group than that in DHEA group （ P 〈 0.001 or P 〈 0.05 ） ,while progesterone level was obviously higher in SF group than that in DHEA group（ P 〈 0.05 ） ;The number of granular cell layer increaseed obviously up to 4 - 8 layers under a microscope in SF group ; apoptosis rate of ovarian granulosa ceils in SF group was significantly lower than that in DHEA group（ P 〈 0.001 ） ; the expression of Bcl - 2 in SF group was significantly higher than that in DHEA group（ P 〈 0. 001 ） ,while the expression of Bax in SF group was significandy lower than that in DHEA group（ P 〈 0. 001 ）. Conclusion The effect of sodium ferulate on improving endocrine condition and reducing the increase range of weight of PCO rats may be relevant to its effect of increasing the expression of Bcl-2 as well as decreasing the expression of Bax in ovarian granulosa ceils.
