摘 要:目的研究神经生长因子(NGF)对体外培养Cajal间质细胞(ICC)的作用及对特征性起搏电流即自发节律性内向电流的影响。方法取CD-l新生小鼠的空肠组织,采用精细快速剥离,取含有肌问丛及肌问丛Cajal间质细胞(ICC-AP)的纵行肌组织块进行培养。12h后加入50ng/mL神经生长因子,3-4d后对照观察、并用膜片钳检测cajal间质细胞的内向电流。结果NGF对培养Cajal间质细胞的生长发育有明显的促进作用,表现为数量增加(每400倍视野细胞数量:NGF 26±8,对照组14±10)、分支增多和网状结构更致密。NGF对培养Cajal间质细胞所产生的自发节律性内向电流与对照组相比表现为电流波形成熟或完整、频率规则而持续、幅度高大而变均衡(电流幅度:NGF179.3pA±29pA,对照组120、8pA±64pA)。结论NGF对于cajal间质细胞的生长发育有明显的促进作用,对特征性起搏电流有加强和稳定作用。Objective To determine the effects of nerve growth factor(NGF) on interstitial cells of Cajal associated with auerbach' s plexus(ICC-AP) culture and to examine the effects of ionic conductance of ICC-AP in response to NGF treated culture. Methods Jejunum was removed from new born CD-1 mice. With sharp dissection, the longitudinal muscle layer containing myenteric plexus was cut into small pieces for culture. After 12 hours of culturing, Tissues were treated with 50 ng/mL NGF in the wells as test group but nothing added in control group. At the day 4, whole-cell currents were obtained from ICC-AP in two groups, respectively. Spontaneously rhythmic inward currents were recorded from whole-cell patch clamping configuration. Results The cells, which were grown from NGF treated cell culture, showed a higher population of ICC-AP (cell amount: NGF 26 ± 8 ,control 14 ± 10) , more branches and denser network than those of control group. The pacemaker current was more regular, consistent and robust in cells from NGF group (amplitude: NGF 179.3 pA ± 29 pA,control 120.8 pA ± 64 pA). Conclusion The growth of ICC-AP can be enhanced by NGF in mouse jejunum explant culture. NGF strongly enhances ICC-AP pacemaker current generation, indicating neural growth factors can regulate the development and signal transduction in ICC-AP.
