摘 要:目的:获取高质量的忍冬基因组DNA。方法:采用改良CTAB法从忍冬叶片中提取DNA,通过琼脂糖凝胶电泳、紫外分光光度法、酶切检测其质量,并与常规CTAB法比较。结果:改良CTAB法所得DNA优于常规CTAB法,电泳条带清晰,完整性好,纯度高,能被完全酶切。结论:改良CTAB法适合忍冬基因组DNA的提取。Objective: To obtain high quality genornic DNA of Lonicera japonica Thunb.. Method: Two different methods,conventional CTAB method and modified CTAB method, were applied to extract genornic DNA from leaves of Lonicera japonica Thunb.. The DNA obtained was analyzed by the means of agarose gel electrophoresis,ultraviolet spectrophotometric method and enzymatic digestion. Result: Higher purity DNA was obtained by modified CTAB method and it can be digested by restriction enzyme completely. Conclusion: The modified CTAB method was favourable for the extraction of genomic DNA from Lonicera japonica Thunb.
