摘 要:用75%乙醇回流法提取出藏药“西藏猫乳(生等)”的有效成分并浓缩精制后,采用HPI—C梯度洗脱法,在选定的色谱条件{DuPont ZORBAX ODS C18色谱柱(4.6mm×25cm),用甲醇(A)~甲酸铵水溶液(pH=3.5,1.0mL·min^-1,B)进行梯度洗脱(甲醇浓度:0~10min,20%~25%;10~20min,25%~40%;20~55min,40%~55%:55~75min,55%~75%;75~85min,75%~90%),检测波长为290nm}下,对藏药“生等”的提取液进行了有效的分离与检测,并建立了“生等”药材的数字化色谱指纹谱(DFPS).所建立的方法能对藏药的产地归属进行有效的分辨.A new separating and determining method was developed for the tibetan medicine Rhamnella gilgitica Mansfeld. et Melch. At first, the refluxing (75% ethanol for 4 h) and concentrating methods were used to extract the effective components of the tibetan medicine. Then, the optimal HPLC conditions were as follows: DuPont ZORBAX ODS C18 Column (4.6 mm×25 cm), methanol(A)-ammonium formate (pH=3.5, 1 mL·min^-1,B) as mobile phase, the gradient elution (solutionA: 0-10 min, 20%-25%, 10-20 min, 25%-40%, 20-55 min, 40%-55%,55-75 min, 55%-75%, 75-85 min, 75%-90%), detection wavelength 290 nm. The accurate and feasible digitized chromatographic fingerprint spectrum(DFPS) of the tibetan medicine was established, which can reflect the real constitution of samples. This new method can be used to distinguish the growing area of the medicine and to control the quality of Rhamnella gilgitica Mansfeld. et Melch samples.
