摘  要:利用RAPD分子标记技术对浙江瓯江溪水性鱼类唇和花进行了种质鉴定和遗传分析,从40个10碱基随机引物中筛选出14个多态性高的引物,分别扩增出130条和127条DNA带,多态位点比率达84.62%和66.93%,平均每个引物扩增多态性带9.3条和9.1条.Shannon多样性指数为0.451 5和0.354 8,Nei s指数为0.303 7和0.239 2.唇和花群体间的平均遗传距离为0.288 0和0.209 6.引物S383、S399、S463和S471扩增的6个特异性标记,可用来区分唇和花.最后应用UPMGA法对它们进行了聚类分析.结果表明唇和花的群体遗传多样性较高,存在较大的遗传变异.RAPD分子标记技术可以方便地应用于属鱼类的种质鉴定.Hemibarbus labeo and H. maculates are important species for freshwater culture. RAPD (random ampli fled polymorphic DNA) markers were applied to the analysis of genetic variation between H. labeo and H. macu lares from Lishui town, zhejiang province. In 14 RAPD primers (screened from 40 primers), each gave rise to 130 and 127 discernible DNA bands, in which 110 (84.62%) and 85 (66.93%) were polymorphic. The average numbers of polymorphic DNA bands amplified by each primer were 9.3 and 9.1. Shannon's diversity index were 0. 451 5 and 0. 354 8 and Nei's genetic diversity index were 0. 303 7 and 0. 239 2. The genetic distances among H. labeo and H. maculates were estimated based on the amount of band sharing were 0. 288 0 and 0. 209 6. Six polymorphic bands amplified from S383, S399, S463 and S471were used to identify them. According to the data, a dendrogram of genetic relationship which were constructed by using UPGMA method showed that the tested species was classified into two cluster groups with the similarity coefficient of 0. 694 in the H. labeo population, while the individuals of H. maculates could also be obtained the same results with the similarity coefficient of 0. 773. All the results of this paper showed that the genetic diversity of H. labeo and H. maculates is high, which means that they have high genetic variation. It further indicated that RAPD technique is a useful tool for the studying of stock identification among the Hemibarbus.
