摘  要:应用末端脱氧核苷酸转移酶介导的生物素化的dUTP切口末端标记(即TUNEL)法和透射电镜(TEM),对人工感染H9N2亚型禽流感病毒蛋鸡的脾脏、胸腺、法氏囊和盲肠扁桃体进行了细胞凋亡动态研究。结果显示,感染蛋鸡脾脏、胸腺、法氏囊中细胞凋亡率升高。接种后3d,脾脏和法氏囊的细胞凋亡率升至最高值分别为6.2%和1.7%,随后下降,但在接种后14d仍显著高于对照组(P<0.05)。胸腺的细胞凋亡率从接种后3~21d均极显著地高于对照组(P<0.01)。而盲肠扁桃体的细胞凋亡率在整个试验期间变化不明显。电镜下凋亡细胞呈典型的形态学特征:细胞染色质固缩并凝结成块,聚集在核膜周边呈新月状或环状小体。结果表明,H9N2亚型禽流感病毒感染蛋鸡能诱导免疫器官的细胞凋亡。Apeptosis of spleen,thymus,bursa of Fabficius and caecum tonsil were demonstrated by the nick end-labelling of DNA in situ (TUNEL) and transmissional electron microscope (TEM). The result showed that apoptosis levels of spleen,thymus and bursa of Fabrieius rose distinctively. Apeptosis rate of spleen,bursa of Fabfieius peaked 6.2% and 1.7% respectively at 3 days after inoculation,then went down,but was very significantly higher than control group (P〈 0.01) .Apeptosis rate of thymus was very significantly higher than control group ( P 〈 0.01) from the 3rd to the 21th after inoculation. Apoptosis rate of caecum consil didn' t significantly changed. TEM showed that some cells had the morphological characteristics of significant apeptosis in the spleen,thymus,bursa of Fabficius and caecum tonsil,and morphological changes were condensation of nuclear ehromatin into dense masses,followed by its margin against the nuclear envelope,and concentration of cytoplasm association with lots of cytoplasmic vacuolization. The results suggested AIr could induce apoptosis of the immune organs in vivo.
