摘  要:目的:观察骨髓间充质干细胞(MSC)对大鼠心脏移植的免疫调节作用。方法:实验共分3组,同种同系移植组(Ⅰ组),供受体均为SD大鼠;同种异系非给药组(Ⅱ组),供体为Wistar大鼠,受体为SD大鼠;同种异系给药组(Ⅲ组),供体为Wistar大鼠,受体为SD大鼠。建立大鼠心脏移植动物模型,贴壁法分离培养大鼠MSC,由Ⅲ组受体颈外静脉注入。观察3组移植后大鼠的存活时间,苏木精-伊红染色观察移植心脏淋巴细胞浸润程度。结果:Ⅰ、Ⅱ、Ⅲ组的平均存活时间分别为(20.8±4.8)d、(6.6±0.9)d、(7.0±1.2)d,Ⅱ组、Ⅲ组与Ⅰ组比较,均差异有统计学意义(P<0.05);但Ⅱ组与Ⅲ组间比较差异无统计学意义。Ⅰ、Ⅱ、Ⅲ组移植心脏病理学分级评分分别为(0.24±0.27)、(3.10±0.55)、(3.00±0.61),Ⅱ组、Ⅲ组与Ⅰ组比较,均差异有统计学意义(P<0.05);但Ⅱ组与Ⅲ组间比较差异无统计学意义。结论:MSC在体外混合淋巴细胞反应中能够抑制T细胞增殖,但是在体内实验中对移植物的存活时间无明显的延长及对移植物的淋巴细胞浸润无明显抑制作用。Objective:To study the immunomodulatory effects of mesenchymal stem cells (MSC) on a model of allogeneic heart transplantation. Method: MSC were obtained from bone marrow aspirates of male SD rats and administered to an allogeneic heart transplantation model. The animals were randomly divided into 3 groups: transplant group: heart transplantation between SD rats; control group., heart from Wistar rats transplant to SD mice; MSCs group., heart from Wistar mice transplant to SD rats, then MSCs were administered to the heart transplantation model. Result: In vivo, MSCs injection did not prolong allograft survival. The xenograft survival of control group and MSC group were (3. 105=0. 55)d and (3.00±0. 61)d, the xenograft survival was not significantly prolonged. Conclusion:Although MSC have immunomodulatory properties in vitro. However, their tolerogenic properties in vivo should be questioned, MSCs injections were ineffective at prolonging allograft survival.
