摘 要:目的:研究罗格列酮(RG)对异丙肾上腺素(ISO)诱导大鼠急性心肌缺血损伤的影响。方法:30只雄性SD大鼠,随机分为3组(对照组、模型组、罗格列酮干预组),用ISO腹腔注射建立急性心肌缺血损伤模型,检测大鼠心功能、血清肌酸激酶(CK)、氧化亚氮(NO)、内皮素(ET)、超氧化物歧化酶(SOD)、心肌丙二醛(MDA)水平及心肌病理组织切片。结果:模型组左心室收缩功能及舒张功能均较对照组明显下降(P<0.05);血清SOD活性、NO水平较对照组降低,CK、ET-1及心肌组织MDA水平较对照组升高(P<0.05);心肌病理损伤明显(P<0.05)。罗格列酮干预组与模型组比较心功能各项指标明显改善(P<0.05),血清CK、ET-1及心肌组织MDA水平降低,SOD活性、NO水平增加(P<0.05),心肌组织病理损伤较模型组减轻(P<0.05)。结论:RG对ISO诱导的大鼠急性心肌损伤具有保护作用,其作用机制可能与其抗氧化及血管内皮细胞保护作用有关。Objective :To investigate the effects of rosiglitazone (RG) on myocardial ischemia injury induced by isoproterenol(ISO) in rats. Methods:Thirty Sprague-Dawley (SD) male rats were randomly divided into three groups (control group, model group and RG intervention group). The rat model of acute myocardial ischemia injury was induced with intraperitoneal injection of ISO. The three groups were evaluated for left ventricle (LV) function, serum levels of creatine kinase ( CK), nitric oxide ( NO), endothelin-1 ( ET-1 ) and superoxide dismutase ( SOD), and myocardial contents of malondialdehyde (MDA) during the experiment. Pathological slides of myocardium from the groups were also studied. Results: Compared with control group, rats in the model group showed significantly poor LV function (P 〈 0.05 ), increased levels of CK, MDA and ET-1, obviously decreased SOD and NO, and evidenced injury of myocardium (P 〈 0.05 ). Compared with model group, the RG intervention group presented with better LV function, lowered levels of CK, MDA and ET-1, as well as elevated SOD and NO ( P 〈 0.05 ) ; Meanwhile, cardiac injury was ameliorated in RG treated rats ( P 〈 0. 05 ). Conclusion: RG appeared protective against myocardial ischemia injury induced by ISO in rats. The underlying mechanism may be associated with anti-oxidation and its protection of vascular endothelial cells.
