高致病性禽流感病毒人工感染小鼠组织病理学观察

摘  要:应用经SPF鸡胚增殖的虎源高致病性禽流感病毒A/Tiger/Harbin/01/2003(H5N1)株(其对MDCK细胞的TCID50为10-7.36/0.05mL),经滴鼻途径人工感染小鼠,无菌采取感染致死的小鼠和对照组小鼠的肺、脑等脏器进行了组织病理学、免疫组化以及透射电镜观察。结果显示:感染致死小鼠以肺脏损害最为明显。光镜观察,肺泡腔不同程度缩小,胞腔内有少量蛋白样浆液渗出,肺泡间隔不同程度增宽,肺内小血管及肺泡间隔的毛细血管扩张、淤血,支气管充血。免疫组化染色发现,在支气管上皮细胞、一些肺泡上皮细胞和浸润的单核细胞胞浆内见到该病毒抗原的阳性染色颗粒。电镜观察,小鼠肺泡扁平上皮细胞表面和肺泡腔中见到短杆型和圆型的流感病毒样粒子。In this study,the HPAIV A/Tiger/Harbin/01/2003(H5N1) originated from tigers was propagated by SPF embryonated hen eggs.TCID50 of virus was 10-736/0.05 mL on MDCK. The mice were inoculated via bronchus,then dead mice were collected to detect by histopathology, immunohistochemistry and transmission electron microscopy. The results showed that the damages of lung from the infected dead mice were the most severe. Sackful infiltrative of macrophage and manipulus protein-like effusion in alveoli was observed under the microscope, lung revealed widening of alveolar space and effusions. It caused telangiectasis and congestion of the small vessels within the lung, and hyperemia in the bronchi. By immunohistochemistry, the positive staining particles of virus were found on the epithelium cells of bronchi and alveolis,and also in the endochylesma of lymphomonocytes. The influenza virion of short-bacillary and spherical type were found within the pavement epithelium cells of alveoli pulmonis in mice by electron microscope. Any other virion had not been isolated from the alveolar epithelium.
