摘 要:研究脑内交感抑制系统中中缝隐核(NRO)至中脑导水管周围灰质腹侧部(VPAG)通路的地茶酚胺递质定量。采用推挽灌流结合高效液相一电化学检测法观察连续电刺激NRO3Omin,vPAG中野上腺素(A)、去甲肾上腺素(NA)释放的变化。结果:刺激NRO10~30min,vPAG灌流液中A含量明显增加(尸<0.O5);刺激NRO10~20min,vPAG中NA含量也增加(P<005)。在上述基础上,于刺激NRO前双侧侧脑室注射5-HT2受体拮抗剂ketanserin(100μg/10μl),给药后电刺激NRO10~3Omin内样品中A、NA含量无明显变化(P>005),ketanserin阻断了刺激NRO引起的A、NA释放增加的效应。结论:NRO至vPAG投射的递质是ANA;5-HT能神经元参与NRO至vPAG的反馈效应,可能由5-HT2受体参与介导。PURPOSE The prsent study was designed to observe dynarnic changes of concentration of catecholamine in the venral periaqueductal grey(vPAG) when nucleus raphe obscurus(NRO) was stimulated.METHODS The experiments were performed in the rats anaesthetized with urethane and alpha-chlo-raose. Push-pull perfusion and HPLC-ECD techniques was used to determin the concentration of catecholamine in vPAG induced by electrical stimulation in NRO.RESULTS There was an increase of adrenaline(A) content in perfusate from vPAG after stimulation was taken for 10 to 30 min P<0.05, and an increase of noradrenaline(NA) for 10 to 20 min (P<0.05). Intracerebroventricle adminstration of the 5-HT2 receptor antagonist ketanserrine blocked the increase of release of A and NA from vPAG induced by stimulation in NRO.CONCLUSIONS The results suggest that effects of stimulating NRO on the vPAG are mediated by A and NA. Serotonergic neurons may be involved in the proection pathway from NRO to vPAG and the receptor invohied may be 5-HT receptor
