摘  要:目的观察可溶性肿瘤坏死因子(TNF)-α受体I重组腺病毒(AdsTNFRI)过继阻断体内TNF-α的生物学活性,探讨其对大鼠肾缺血再灌注的治疗作用。方法雄性SD大鼠随机分成4组:假手术组、肾缺血再灌注模型组、肾缺血再灌注-sTNFRI组和肾缺血再灌注-Ad-LacZ组。缺血2h后,再灌注24h。取各组大鼠尾静脉血,制备血清。测定各组血清中TNF-α、sTNFRI、抗氧化酶T-SOD、CAT及脂质过氧化物丙二醛(MDA)水平。结果缺血再灌注大鼠体内抗氧化酶T-SOD、CAT较假手术组显著降低,TGF-α、MDA显著增高。可溶性TNF-α受体I重组腺病毒(Ad-sTNFRI)过继给缺血再灌注大鼠体内后,T-SOD、CAT、可溶性TNF-α受体I(sTNFR-I)显著增高,TGF-α、MDA显著降低。结论AdsTNFRI可中和缺血再灌注大鼠外周血中TNF-α水平,并可增强其抗氧化能力,降低MDA水平,抑制缺血再灌注大鼠体内氧化应激水平,对大鼠缺血再灌注有一定的治疗作用。Objective To explore the protectiive effects by injecting recombinant adenovirus encoded soluble TNF-α receptor-I into renal ischemia-reperfusion rats. Methods SD rats were randomly di- vided into 4 groups with 15 animals in each group. Group A:sham operation group;Group B:renal ischemi- a- reperfusion group ; Group C : renal ischemia- reperfusion group with injection of recombinant adenovirus encoded soluble TNF-α receptor-I (sTNFR I ) gene;Group D:renal ischemia- reperfusion group with in- jection of recombinant adenovirus encoded LacZ gene. Rats in group B, group C and group D were reper- fused for 24 h,followed by 2 h ischemia. The serum was collected and the levels of TNF-α,sTNFR I ,total superoxide dismutase (T-SOD) ,catalase (CAT) and malonaldehyde (MDA) were determined. Results The levels of T-SOD and CAT in group C were lower than those in group A, and the levels of TGF-α and MDA were higher significantly ( P 〈 0.01 ). The levels of T-SOD, CAT and sTNFR I were strikingly heightened when compared with those of group D, and the levels of TGF-α and MDA were declined markedly. Conclusion Recombinant adenovirus encoded soluble TNF-α receptor-I (AdsTNFR I ) could neutralize TGF-α,increase the capability of anti-oxidization, reduce the expression of MDA, and supress oxidative stress in renal isehemia-reperfusion rats.
