加味四逆散对慢性心理应激抑郁症大鼠海马损伤的保护作用

摘　　要：目的观察加味四逆散对慢性心理应激抑郁症大鼠海马损伤的保护作用。方法采用多种应激源制作慢性心理应激抑郁症大鼠模型。30只Wistar大鼠分为正常组、模型组、中药组。观察各组大鼠海马神经元凋亡率,测定海马脑源性神经营养因子(BDNF)及其受体TrkB蛋白含量及神经元内Ca2+浓度。结果模型组大鼠海马神经元凋亡率较正常组增加,海马BDNF及其受体TrkB蛋白表达降低,神经元内Ca2+浓度升高(P<0.05或P<0.01)。中药组可降低海马神经元凋亡率,增加BDNF与TrkB蛋白表达,降低神经元内Ca2+浓度。结论加味四逆散可能通过上调海马BDNF的表达,降低胞内Ca2+,实现抗抑郁的作用。Objective To investigate the protective effect of Modified Sini Powder （Modified powder for treating cold limbs） on hippocampus injury in chronic stressed depression rats. Methods Multistressors were adopted to establish the rat model of chronic stressed depression. Totally 30 male Wistar rats were randomly divided into 3 groups： the control group, model group, and Modified Sini Powder group. Hippocampal neuron apoptosis rate,content of brain-derived neurotrophic factor （BDNF） and its receptor TrkB protein, and Ca^2＋ concentration in neurons of each group were observed. Results Stressed rats in the model group had significantly higher hippocampal neuron apoptosis rate than those in the control group, BDNF and its receptor TrkB were decreased in expression, and Ca^2＋ concentration in neurons was significantly higher （P〈0.05 or P〈0.01）. In the Modified Sini Powder group, hippocampal neuron apoptosis rate was lowered, BDNF and TrkB protein expression increased, and Ca^2＋ concentration in neurons decreased, the difference was significant in comparison with those of the model group （P〈0.05 or P〈0.01）. Conclusion Modified Sini Powder can improve depression by up-regulating the expression of BDNF in hippocampal neurons and decreasing intracellular Ca^2＋ concentration.
