摘  要:目的研究参杞合剂(SQ)对人鼻咽癌细胞CNE的凋亡及转移的影响,探讨其对人类肿瘤细胞作用的机制。方法以四氮唑盐(MTT)法计算抑瘤率;琼脂糖凝胶电泳观察凋亡细胞形成的DNA条带;流式细胞术检测Fas的表达;逆转录聚合酶联反应(RT-PCR)检测SQ对半胱氨酸蛋白酶3(caspase-3)表达的影响。结果SQ对CNE细胞具有明显的直接杀伤作用;琼脂糖凝胶电泳可以检测到凋亡特征性条带;Fas表达上调;caspase-3表达增加。结论SQ可诱导人鼻咽癌细胞凋亡,可能与Fas/FasL介导的死亡受体途径有关。Objective To investigate the effect of Shenqi Mixture(Mixture of Radix Codonopsis and Fructus Lycii) on CNE cell apoptosis and metastasis of nasopharyngeal cancer to study its mechanism for human tumor cells. Methods The methyl thiazolyl terrazolium (MTT) was applied to calculate the tumor inhibition rate. The agarose gel electrophoresis was adopted to observe the DNA band formed by apoptosis cells. The flow cytometry (FCM) was employed to test the Fas expression. The reverse transcription polymerase chain reaction (RT-PCR) was used to determine the effect of Shenqi Mixture on the expression of caspase-3. Results Shenqi Mixture could directly kill CNE cells. The apoptosis band was seen in the agarose gel electrophoresis. The increased expression of Fas and caspase-3 was observed. Conclusion Shenqi Mixture can induce the cell apoptosis of human nasopharyngeal cancer possibly through the Fas/FasL pathway.
