3种测定血清α-L-岩藻糖苷酶试剂的评价

摘　　要：目的对浙江东瓯、德国Autec、南京威特曼3种AFU试剂的评价，为选择AFU试剂提供依据。方法比较3种试剂的精密度、准确性以及抗坏血酸、肝素钠、血红蛋白、胆红素、甘油三酯对测AFU的干扰能力。结果3种试剂的批内、日间变异系数均小于3％，总变异系数均小于5％，彼此相关系数为0．9159、0．9288、0．9824，回收率为98．3％～101．1％，线性达224u／L，维生素C大于或等于0．238mg／mL、Hb≥0．5g／L对AFU产生负干扰，肝素钠对AFU≥10IU／mL产生正干扰，威特曼试剂受这些干扰物的影响较其他两种试剂小；胆红素小于或等于135．7μmol／L对AFU没有干扰，甘油三酯小于或等于6．60mmol／L时，东瓯和Autec试剂不受干扰，而甘油三酯大于或等于4．80mmol／L时，威特曼试剂受影响较大。结论3种试剂在抗干扰能力上有明显差异，正常血清标本均能获得满意结果。Objective To evaluate the three reagents （Dong Ou,Autec,Whiteman）measuring AFU,and provide a basis in selection of AFU reagent. Methods Compared of the precision,accuracy and the interference of ascorbic acid hepcorrin sodium, hemoglobin,bilirubin and triglyceride toward AFU the three reagents. Results It was proved that in the three reagents the within-run variance and day to day variance were all less 3%, the total variance was less than 5 %, they correlated well eath other（r= 0. 915 9, 0. 928 8,0. 982 4）, Recoveries were 98. 3%-101.1%, and the linearity was up to 224u/L,samples with Vitamin C≥0. 238mg/ mL, hemoglobin≥0.5g/L showed lower results,samples with heparin sodium ≥10IU/mL showed higher results Whiteman reagent had smaller effect by this interference compared with the other two reagents. It also proved that Bilirubin 4135.7μmol/L dit not interfere AFU. When triglyceride was 46.6mmol/L,Autec and Dong Ou reagents did not effect,Whereas triglyceride was ≥4.8 mmol/L,Whiteman affected much. Conclusion The evaluation approve that the anti-jamming capacities of the three reagents have obvious difference, but the regular serum sample all can get satisfyactory result.
