摘  要:目的:探讨TNF-α对胸苷磷酸化酶(TP酶)的调节及其与5-FU前体药化疗敏感性的关系.方法:应用TNF-α诱导肿瘤细胞中的TP酶,ELISA法检测肿瘤组织中TP酶蛋白水平,采用MTT法检测药物敏感性变化.结果:大肠癌细胞株Lovo和SW620中未检测到TP酶,乳腺癌细胞株MCF-7中的TP酶含量为(30.1±1.4)μg/g,高于MCF/ADR的TP酶(18.8±0.4)μg/g.TNF-α上调MCF-7和MCF/ADR中TP酶表达呈时间和剂量依赖性.耐药株MCF/ADR中TP酶上调高达(28.4±1.2)μg/g,未达非耐药细胞株水平.MCF-7细胞中TP酶上调1.27倍,对氟铁龙的IC50降低了1.33倍,有统计学意义.结论:TNF-α能剂量-时间依赖性上调肿瘤细胞的TP酶,TNF-α为5-FU前体药化疗增效.AIM: To explore the regulative action of TNF-α on thymidine phosphorylase (TP) and its effect on the sensitivity of prodrug to 5-fluorouracil ( 5-FU ) in cancer cell lines. METHODS: The TP level of tumor cells was mearsured by ELISA. And then the MTY assay was done for detecting the drug sensitivity of the cells treated with small concentration of TNF-α. RESULTS: The TP in Lovo or SW620 was not displayed, and it was significantly higher in MCF-7 [ ( 30. 1 ± 1.4 ) μg/g ] than in MCF/ADR[ (18.8 ±0.4)μg/g]. TNF-α caused a concentration and time dependent increase in the level of TP. The TP level of MCF-7 displayed a 1.27-fold increase, while its IC50 displayed a 1.33-fold decrease. CONCLUSION: TNF-α can cause a concertration and time dependent increase in the level of TP and enhance the sensitivity of MCF-7 to 5-FU.
