摘  要:目的:筛选红车轴草中异黄酮的最佳提取条件,研究大孔树脂富集、纯化总异黄酮的工艺条件与参数。方法:以总异黄酮的含量和重量为指标,采用正交试验筛选红车轴草的最佳提取方法和吸附条件。结果:红车轴草的最佳提取工艺为20倍量的70%乙醇,提取2次,每次1.5h,洗脱条件为以4倍量柱体的水和30%乙醇洗去杂质,再以4倍量的90%乙醇洗脱,收集总异黄酮的含量和得率最高;最佳上柱条件为药液浓度1∶4,径高比1∶20,药液洗脱流速20mL·min-1。结论:采用此法可较好地富集、纯化红车轴草中的总异黄酮。Objective: To screen the best method of extraction of total isoflavones from Trifolium pretense L.. To study the technological parameters of the purification process of total isoflavones from Trifolium pretense L.. Methods: Use the content and weight of total isoflavones as maker to screen the best method of extraction and obsorption parameters by orthogonal design. Results:The best method of extraction were extracting the Trifolium pretense L two times with 20BV 70% ethanol and the extraction time is 1.5 hour. The best desorption condition is washing the resin volume with 4BV 90% ethanol. The result showed that extract solution at a concentration 1:4, ratio of column diameter to column height 1:20, flow rate of 20mL·min^-1 as the optimal condition. Conclusion: The purification of total isoflavones from Trifolium pratense L is reliable and efficacious.
