摘  要:目的采用实时荧光定量PCR技术同时检测新生儿黄疸患者外周血白细胞和血浆中巨细胞病毒,并分析两者之间的相关性。方法采用0.8%氯化胺(NH4Cl)裂解红细胞法提取新生儿黄疸患者外周血白细胞,然后进行实时荧光定量PCR检测,同时检测血浆中巨细胞病毒。结果79例标本中白细胞和血浆均检出巨细胞病毒7例,阳性率为8.86%;白细胞巨细胞病毒定量平均值为4.16(对数值);血浆平均值为2.37(对数值),经相关性分析,两者呈正相关(r=0.78)。结论荧光定量PCR检测巨细胞病毒快捷、简便、高效;提取白细胞扩增和血浆水平高度相关。Objective To detect simultaneously peripheral blood leucocyte and plasma cytomegalovirus in patients with neonataljaundice with real-time fluorescence quantitive PCR (FQ-PCR) and analyze their correlation. Methods After the erythrocytes were lysed with ammonium chloride (0. 8%, W/V), peripheral blood leucocytes were abstracted. FQ-PCR was applied to detecting simultaneously peripheral blood leucocyte and plasma cytomegalovirus in patients with neonataljaundice. Results In 79 cases of samples, 7 cases were detected in peripheral blood leucocyte and plasma respectively, and the positive rate was 8.86%. The quantitation mean value (logarithm) of leucocytes and plasma was 4. 16 and 2.37 respectively, Correlation analysis showed they were positively correlative (r= 0.78). Conclusion FQ-PCR is a rapid and reliable method for cytomegalovirus detection. The level of peripheral blood leucocyte cytomegalovirus is closely correlative to that of plasma cytomegalovirus.
