摘  要:【目的】探讨吗啡依赖和戒断小鼠海马金属硫蛋白(metallothionein,MT)MT1、MT2 mRNA表达的变化。【方法】剂量递增法皮下注射吗啡建立吗啡依赖小鼠模型,腹腔注射盐酸纳络酮诱发戒断症状。根据小鼠戒断反应中出现的跳跃次数、体重下降等指标评定戒断反应的强度。采用半定量逆转录-聚合酶链反应(RT-PCR)方法,观察实验小鼠海马MT1、MT2 mRNA的变化情况。【结果】吗啡依赖组MT1、MT2 mRNA表达水平高于对照组和吗啡戒断组(P<0.05),吗啡戒断组MT1、MT2 mRNA表达水平高于对照组(P<0.05)。【结论】吗啡依赖小鼠海马MT1、MT2 mRNA表达增加,纳络酮戒断能够抑制吗啡依赖引起的MT1、MT2表达增加,表明海马MT的表达变化参与了吗啡依赖和戒断过程。[Objective] To investigate the effect of morphine dependence and withdrawal on the expression of MT1 mRNA and MT2 mRNA in hippocampus of mice. [Methods] Morphine was subcutaneously injected into the mice with dose-increasing method to establish morphine dependence model and withdrawal syndromes were precipitated by naloxone. The intensity of withdrawal syndromes was evaluated according to indices, such as the number of jumping, the weight loss, et al. Semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) method was used to detect the expression of MT1 mRNA and MT2 mRNA in the hippocampus of experimental mice. [Results] Compared with control group and morphine withdrawal group, the expressions of MT1 mRNA and MT2 mRNA were up-regulated in morphine dependent group (P 〈 0.05), the expression levels of MT1 mRNA and MT2 mRNA were higher in morphine withdrawal group than in control group (P 〈 0.05) ; [Conclusions] The expression of MT1 mRNA and MT2 mRNA in hippocampus was up-regulated in morphine dependent group, and this phenomenon can be inhibited by naloxone, indicating that the changes of the expression of MT mRNA take part in the course of morphine dependence and withdrawal.
