摘 要:目的观察血库常规保存的全血不同保存时间血浆中性粒细胞肽(HNP)和中性粒细胞弹性蛋白酶(NE)含量的动态变化,探讨不同保存时间对多形核中性粒细胞(PMN)稳定性的影响。方法使用ELISA夹心法,对36名健康献血者新鲜血浆和血库常规保存的9份全血不同保存时间血浆HNP和NE含量进行了定量测定。结果36名健康献血者血浆HNP和NE含量分别为(46±48)ng/ml和(35±28)ng/ml;常规保存的9份全血随保存时间的延长血浆HNP和NE含量逐渐升高,保存到10天时的全血血浆HNP浓度和保存到15天时的全血血浆NE浓度(77±43)ng/ml与第1天相比升高显著(P<0.01),且HNP和NE之间的含量变化呈正相关(r=0.96,P<0.01)。结论库存全血血浆中HNP和NE随保存时间延长含量增加,可能是PMN胞浆成分逐渐进入血浆所致。Objective To dynamic observe plasma human neutrophil peptide(HNP) and neutrophil elastase(NE) content and stability of PMN in different time of preservation. Methods According to the ELISA method, the HNP and NE content of 36 healthy plasma and 9 plasma of the blood bank conventional preservation were determined. Results The HNP contents of 36 healthy plasma were (46±48) ng/ml. The NE contents of 36 healthy plasma were (35±28) ng/ml. The HNP and NE contents of 9 plasma were increase along with the increased storage time for the blood bank conventional preservation. When saving to 10 days, there was significant difference in lIMP content between the first day[(45±42) ng/ml] and the 10th day[(98±61) ng/ml] (P〈0. 01). When saving to 15 days, there was significant difference in NE content between the first day[(34±27) ng/ml and the 15th day[(77 ±43) ng/ml](P〈0. 01). HNP presented positive related to the NE(r= 0. 96,P〈 0. 01). Conclusion For reason that the PMN cytoplasmic components soak into plasma gradually, the plasma HNP and NE content is increase along with the increased storage time for the blood bank conventional preservation.
